Letrozole: 7 things you should know
If you’re experiencing certain side effects of letrozole, you may want to avoid foods that can worsen them. To put one example into perspective, the Royal Osteoporosis Society recommends limiting your coffee intake to 4 cups per day if you have risk factors for osteoporosis. Healthcare professionals generally recommend continuing with your normal diet while taking letrozole, and you can take this medication with or without food. The results for the study comparing letrozole to aminoglutethimide (AR/BC3), with a minimum follow-up of 9 months, are shown in Table 18 (Unadjusted analyses are used). Letrozole was not mutagenic in in vitro tests (Ames and E.coli bacterial tests) but was observed to be a potential clastogen in in vitro assays steroids (CHO K1 and CCL 61 Chinese hamster ovary cells).

What are some things I need to know or do while I take this drug?
- Advise females of reproductive potential to use effective contraception during treatment with letrozole and for at least 3 weeks after the last dose.

The following information includes only the average doses of this medicine. If your dose is different, do not change it unless your doctor tells you to do so. Although certain medicines should not be used together at all, in other cases two different medicines may be used together even if an interaction might occur.

Letrozole was teratogenic to rats at a dose of 0.03 mg/kg (approximately 0.01 times the maximum recommended human dose on a mg/m 2 basis) and caused fetal domed head and cervical/centrum vertebral fusion. Letrozole lowers the levels of the female sex hormone oestrogen in the body. These breast cancers are called hormone sensitive or hormone receptor positive. Letrozole can stop or slow the growth of these cancers.

If side effects cannot be managed, your doctor may suggest a different type of hormonal therapy. Other cancer treatments may cause different side effects. If you are also having other cancer treatment, you may have other side effects. If you have not been through the menopause, you may have other types of hormone therapy.
In the final updated analysis, conducted at a median follow-up of 62 months, the median treatment duration for letrozole was 60 months. Seventy-one (71%) percent of patients were treated for at least 3 years and 58% of patients completed at least 4.5 years of extended adjuvant treatment. The treatment should be discontinued at tumor relapse see Clinical Studies (14.2). Letrozole Tablets, USP are indicated for the adjuvant treatment of postmenopausal women with hormone receptor positive early breast cancer.
